We just spent a lot of time trouble shooting an issue where 2008 R2 and 2008 SP2 guests on VMWare would fail to backup with error: Failed to get access to hard disk [share storage]/guest.vmdk. This seemed to only effect servers that were provishened via v-sphere. Servers with these OS that were converted with VM Conversion Tools did not seem to have this problem.

VMTools installation by default installs VMWare shadow copy service. If this is installed, then backups will fail. Unfortunately, the only way to tell if the option is on, is to re-run the VMTools installation in interactive mode while connected to a console session. Modify the installation and remove Volume Shadow Copy option. This should allow acronis to start backing up these VM's. If you have not installed VM Tools yet, then you may want to run a custom install and de-select this option. There is no GUI option in the guest or in v-sphere to tell if this option is enabled.
