Need to convert external storage drive from FAT32 to NTFS

I'm a newby so silly question coming up.
I need to transfer a 12GB pst file from Outlook on an existing desktop machine to Outlook on a new desktop. I assumed that my 1TB Buffalo external drive would be ideal as I use it every evening to back up my laptop on a scheduled task. Problem is, a dialog keeps telling me that there's insufficient space on the disk (which is nonsense as there's 180Gb free).
The email provider pointed out that if the external disk is in FAT32 format, then it's limited to file sizes of 4 gigs. If I manage to convert the external drive to NTFS, will this have a detrimentalo effect on Acronis True Image 2012?
