Can't change drive letter on USB stick!

It is hard to say. I assume you have attempted to change the drive letter using Disk Management Ap. The only thing I can think of is that there could be a permissions problem; you could try logging in a Administrator.

Did a but if research and looks like this can happen if there is an open file on the USB drive. Same sort of problem when you try to eject a USB drive and OS thinks there is still a file open. Sometimes having Explorer open on the drive can trick OS into thinking there is an open file (or so it seems so me). 

Try restarting the computer and see if the problem persists.

There are more involved ways to find out if anything is keeping files open on the USB drive, but it's far simpler to just shutdown and disconnect.

Can you not change the drive letter at all? Or only not to a specific letter?

You could also see if you force the drive letter to change and see what happens. I.e. disconnect problematic drive, insert another usb drive, connect the problematic drive again. 

Some form of screenshot from the disk manager could also help to suggest more stuff to do.
