Backup Partitions & Rescue Media

I am using ATI2014 on a Windows 8.1 64bit desktop. The computer is only a week old.
I'm running a FULL backup weekly. When performing the backup, the GUI shows 5 partitions on the disk (ESP, DIAGS, WINRETOOLS, PBR Image, and OS(C)). I'm not sure what all these partitions are for and that's probably not important. All partitions have the green check mark except DIAGS. Should I manually check this box to include it in the backup?
Also, I have created rescue media on a USB flash drive and tested it to make sure it boots the computer and that it sees my backups (created on an external HD). Should I create the backups using the rescue media or is it acceptable to simply create the backup from within the Windows environment?

Thank you for those answers. I'd like to follow-up with another question.
I have completed a clean install of ATI2014 17.0.6673 (uninstalled using Programs and Features > Acronis Clean-Up Utility > rebooted).
The USB flash drive I have been using as rescue media was made using the ATI2013 rescue media builder.
I formatted the USB flash drive by right-clicking it and choosing FORMAT. I understand that the rescue media should be made with the current product and version and pr0eceded to do so from within ATI2014. At some point, the installation stopped and said something to the effect that the USB flash drive must be formatted with the FAT file structure. It seems that Windows 8.1 does not offer this option, only exFAT which ATI2014 doesn't like.
Are there any options for me so I can use the USB flash drive for rescue or should I use a CD to install the rescue software?

It's a little difficult to deal with repartitioning USB flash drives in Windows. There are a number of free disk partitioning programs available. MiniTool Partition Wizard Home Edition is one. Use it to delete the partition and create a new partition (no bigger that 32 GB) and format it FAT32. You could also use diskpart from a command line, but a partitioning program is much safer.

The Minitool Partition Wizard Home Edition did the trick (once I figured out you had to hit the APPLY button to start the format process). The USB flash drive is now FAT32, the rescue files have been written to it, it boots into the rescue environment, and my backups are visible.

One easy way to make sure all partitons are included in your backup is to use the option "Switch to disk mode" as illustrated in signature link #2 below and the very first picture.
