NTFS vs FAT32

I am currently using True Image 2011 Trial version. I plan to buy the package soon.
My internal boot drive is NTFS, as are two of my external drives. A new external drive which I plan to use with true image is FAT32. I am wondering if I should move the data somewhere else temporarily and then reformat the drive to NTFS.
